A Prospective Evaluation of MicroRNAs as a Meniere’s Disease Biomarker

Abstract:
Background: Meniere’s Disease (MD) is a debilitating inner ear disorder characterized by vertigo, sensorineural hearing loss, and tinnitus. The clinical presentation of MD is variable, and more precise diagnostic tools are needed. Cluster analysis of perilymph has suggested there are at least 5 different categories of MD and multiple different genes implicated. At this point in time, the diagnosis is made purely on the clinical presence of sensorineural hearing loss, attacks of vertigo and fluctuating aural symptoms. Previous studies performed in our lab have identified microRNAs differentially expressed in the perilymph and serum of patients with active MD.
Methods: Prospective single-center study. Patients with active, bilateral MD and age matched controls were enrolled. Evaluation of serum miRNA expression was performed using microarray and quantitative PCR. Presence of concurrent autoimmune disease or migraines was noted.
Results: miRNA expression in serum could be correlated to the diagnosis of Meniere disease. There was a high level of variance in the expression level of microRNA-1299.
Conclusions: Development of reliable biomarkers for Meniere’s disease will lead to better classification of subgroups of Meniere disease, leading to the development of novel treatments.

I grew up in Chandler, Arizona and moved to Kansas City as a new college graduate to work as a laboratory manager.
